Well I saw this yesterday, and it was a really fucked up movie--not surprisingly. I made the mistake of watching both this and Inland Empire for the first time, while stoned, and my brain still feels like scrambled mush.
Even though it's written and directed by Charlie Kaufman, Synecdoche seems very Lynch-ian actually (ie a complete mindfuck) and while being extremely fractured and fucked up, it seems to have more emotion than Lynch's flicks. I found myself a bit choked up, even though I was also very lost.
Like another Kaufman movie Adaptation, Synecoche is metafictional to the point of sort of.. curling in on itself -- Though this time it gets seriously out of hand. I highly recommend this to anyone who's a fan of Kaufman or Lynch. It's refreshing to see relatively mainstream writers/directors attempt to shatter boundaries in storytelling like this.

I'm glad others are interested in this movie. Some spoiler-type discussion would be in order once more of you have seen it.
This movie is difficult to wrap your head around -- very philosophical and dense.
I think Kaufman is similar to Lynch in their general weirdness. But I don't think Kaufman is usually weird just for the sake of it... His weirdness is part of a plot device usually -- The stories rely heavily on these weird aspects, which some might consider to be gimmicks. But his forte is using those bizarre concepts to expose the frailty and humanity of his characters... While probing the viewer and his/her emotions in a way that's nearly impossible with conventional film.

Its not as deep as you guys seem to think it is. Its certainly not lynch. While it had some beautiful moments, its more a bunch of random stuff strung together with a semi-reasonable plot so that it appears to have more meaning than it really does. That in itself is practically what david lynch does, however this movie doesn't have any atmosphere.

I thought it was pretty good, and that's the truth! I'm siding with Taxidermist and erasing his "random stuff" statement. The camera always goes to the person talking, not just anyone, and the twists were more alienating that random, that's the point.
Worth watching again for detailed sets and character checklist, though I probably won't. 3.5/5

I think it has a dead atmosphere, the kind of disassociated atmosphere which reminds me of taking LSD; fractured & neutral, huge & silent.

just saw this... really terrific movie. might rival eternal sunshine now as my favorite kaufman movie
very complex story, visually interesting, and philip seymore hoffman was ace as always
